RESEARCH SUMMARY

Plant–derived metabolic agents have a recognizable proportion of the currently available drugs for various treatments from microbial infection to cancer. Thanks to ethnobotany, pharmacognosy, and ethnopharmacological approaches. Basd on these approacheis, we managed to conduct studies that aimed to verify claims made on safety and efficacy of many reputable plant species and to establish several bioassay models such as cyclooxygenase, lipoxygenase (antiinflammatory) and  acetylcholinesterase (central nervious system disorders) inhibitory screening models.We have sucessfully isoalted a number of bioactive agents to be developed for further applications such as: Anolignan B, Termilignan, Niloticane and Cycloartane among others.

Currently, we are focusing on plants from marine ecosystems in particular our magical mangroves.  The collective noun mangrove designates an inter-tidal wetland ecosystem formed by a very special association of animals and plants in the coastal areas and river estuaries throughout the low lying tropical and sub-tropical latitudes. Mangroves are highly productive ecosystem with various important economic and environmental functions. They exist under stressful conditions such as violent environments, high concentration of moisture, high and low tides of water, and abundant living microorganisms and insects. Due to this special growth environment, mangroves produce diverse group of metabolic substances with wide range of biological activities.

Although the chemical constituents of most mangrove plants still have not been studied extensively, our investigations have led so far to the discovery of different compounds and/or fractions with prospective medicinal values.

Our current project is focused on natural products with anti-rheumatoid arthritis properties. Rheumatoid arthritis (RA) is an autoimmune disease characterized by chronic inflammation of joints leading to progressive and irreversible joint destruction. This partially caused by an aggressive invasion of synovial tissue, called pannus, to local articular structure. The pannus is characterized by a synovial hyperplasia composed mainly of fibroblast-like synoviocytes (FLSs) combined with a massive infiltration of lymphocytes and macrophages. In this project we are looking for molecules that supress the up normal proliferation of fibroblast-like synoviocytes.

 

Apart from the mangrove plants, we are also looking at the endophytes as a rich source of bioactive entities. Endophytes are microorganisms that live in the intercellular spaces of plant tissue with no apparent damage to their host. Endophytes represent a huge diversity of microbial adaptations that have developed in special sequestered environment. This could therefore, act as a selection pressure for the development of novel metabolic pathways which may lead to exquisite end products. Thanks to the endophytic fungus Taxomyces andreana and its host plant Taxus brevifolia for the production of the taxol, the famous anti-cancer compound. Endophytes therefore, are viewed as an outstanding source of bioactive natural products that need to be fully discovered. We have isolated a number of endophytic bacterial strains that produce potential antimicrobial substances.
